Ghaderi, Darius; Springer, Stevan A. Humans lack the common mammalian cell surface molecule N-glycolylneuraminic acid Neu 5 Gc due to a CMAH gene inactivation, which occurred approximately three million years ago. Modern humans produce antibodies specific for Neu 5 Gc. We hypothesized that anti- Neu 5 Gc antibodies could enter the female reproductive tract and target Neu 5 Gc -positive sperm or fetal tissues, reducing reproductive compatibility.
N-glycolyl groups of nonhuman chondroitin sulfates survive in ancient fossils. Biosynthesis of the common mammalian sialic acid N - glycolylneuraminic acid Neu 5 Gc was lost during human evolution due to inactivation of the CMAH gene, possibly expediting divergence of the Homo lineage, due to a partial fertility barrier. Neu 5 Gc catabolism generates N -glycolylhexosamines, which are potential precursors for glycoconjugate biosynthesis.
We carried out metabolic labeling experiments and studies of mice with human-like Neu 5 Gc deficiency to show that Neu 5 Gc degradation is the metabolic source of UDP-GlcNGc and UDP-GalNGc and the latter allows an unexpectedly selective incorporation of N -glycolyl groups into chondroitin sulfate CS over other potential glycoconjugate products.
Partially N -glycolylated-CS was chemically synthesized as a standard for mass spectrometry to confirm its natural occurrence. Unlike the case with Neu 5 Gc , N -glycolyl-CS was also stable enough to be detectable in animal fossils as old as 4 My. This work opens the door for investigating the biological and immunological significance of this glycosaminoglycan modification and for an "ancient glycans" approach to dating of Neu 5 Gc loss during the evolution of Homo. The activation of sialic acids Sia to CMP-Sia is a prerequisite for the de novo synthesis of sialoglycoconjugates.
In vertebrates, CSS has been cloned from human, mouse, and rainbow trout, and the crystal structure has been resolved for the mouse enzyme. The mouse and rainbow trout enzyme have been compared with respect to substrate specificity, demonstrating that the mouse enzyme exhibits a pronounced specificity for N-acetylneuraminic acid Neu5Ac , while the rainbow trout CSS is equally active with either of three Sia species, Neu5Ac, N-glycolylneuraminic acid Neu 5 Gc , and deaminoneuraminic acid KDN. However, molecular details that explain the pronounced substrate specificities are unknown.
Understanding the catalytic mechanisms of these enzymes is of major importance, since CSSs play crucial roles in cellular sialylation patterns and thus are potential drug targets in a number of pathophysiological situations. The availability of the cDNAs and the obtained structural data enable rational approaches; however, these efforts are limited by the lack of a reliable high-throughput assay system.
Here we describe a new assay system that allows product quantification in a reduced nicotinamide adenine dinucleotide NADH -dependent color reaction. Consumption of NADH can be photometrically monitored on a microtiter plate reader for a number of test samples at the same time. Full Text Available Neisseria gonorrhoeae deploys a novel immune evasion strategy wherein the lacto-N-neotetraose LNnT structure of lipooligosaccharide LOS is capped by the bacterial sialyltransferase, using host cytidine-5'-monophosphate CMP-activated forms of the nine-carbon nonulosonate NulO sugar N-acetyl-neuraminic acid Neu5Ac, a sialic acid Sia abundant in humans.
This allows evasion of complement-mediated killing by recruiting factor H FH, an inhibitor of the alternative complement pathway, and by limiting classical pathway activation "serum-resistance". While serum-resistance mediated by Neu5Ac was associated with classical pathway inhibition decreased IgG binding and C4 deposition, Leg5Ac7Ac and Neu5Ac9Az incorporation did not inhibit the classical pathway.
Surprisingly, despite near-maximal FH binding in this instance, the alternative pathway was not regulated and factor Bb remained associated with bacteria. Full Text Available We examined whether deficiency of the GGTA1 gene in pigs altered the expression of several glycosyltransferase genes.
Enzyme-linked lectin assays indicated that there were also more sialic acid -containing glycoconjugate epitopes in GalT KO livers than in controls. This indicates that Neu 5 Gc may be a critical xenoantigen. Chemoselective synthesis of sialic acid 1,7-lactones. The chemoselective synthesis of the 1,7-lactones of N-acetylneuraminic acid , N-glycolylneuraminic acid , and 3-deoxy-d-glycero-d-galacto-nononic acid is accomplished in two steps: The instability of the 1,7-lactones to protic solvents has been also evidenced together with the rationalization of the mechanism of their formation under acylation conditions.
The results permit to dispose of authentic 1,7-sialolactones to be used as reference standards and of a procedure useful for the preparation of their isotopologues to be used as inner standards in improved analytical procedures for the gas liquid chromatography-mass spectrometry GLC-MS analysis of 1,7-sialolactones in biological media. Several samples were collected during the production of the antibodies in hollow-fibre reactors.
The heavy chain, but not the light chain, of the two different chimeric IgG1 antibodies is glycosylated. Structural analysis of the enzymically released N-linked carbohydrate chains by 1H-NMR spectroscopy, as well as by chromatographic profiling, demonstrated that the collection of N-glycans comprises a small amount of monoantennary, and for the greater part diantennary structures. Analysis of the different production batches demonstrates that the structures of the N-linked carbohydrate chains are identical in the two.
Structural characterization of neutral and acidic oligosaccharides in the milks of strepsirrhine primates: The structures of milk oligosaccharides were characterized for four strepsirrhine primates to examine the extent to which they resemble milk oligosaccharides in other primates.
Neutral and acidic oligosaccharides were isolated from milk of the greater galago Galagidae: Otolemur crassicaudatus , aye-aye Daubentoniidae: Daubentonia madagascariensis , Coquerel's sifaka Indriidae: Propithecus coquereli and mongoose lemur Lemuridae: Eulemur mongoz , and their chemical structures were characterized by 1 H-NMR spectroscopy.
The oligosaccharide patterns observed among strepsirrhines did not appear to correlate to phylogeny, sociality or pattern of infant care. Both type I and type II neutral oligosaccharides were found in the milk of the aye-aye, but type II predominate over type I. Only type II oligosaccharides were identified in other strepsirrhine milks. Oligosaccharides containing the Lewis x epitope were found in aye-aye and mongoose lemur milk. The finding of a variety of neutral and acidic oligosaccharides in the milks of strepsirrhines, as previously reported for haplorhines, suggests that such constituents are ancient rather than derived features, and are as characteristic of primate lactation is the classic disaccharide, lactose.
Exogenous incorporation of neugc-rich mucin augments n-glycolyl sialic acid content and promotes malignant phenotype in mouse tumor cell lines. Full Text Available Abstract Background Carbohydrates embedded in the plasma membrane are one of the main actors involved in the communication of cells with the microenvironment. Neuraminic sialic acids are glycocalyx sugars that play important roles in the modulation of malignant cell behaviour.
N-glycolylneuraminic acid NeuGc is synthesized by the cytidine monophospho-N-acetylneuraminic acid hydroxylase CMAH, an enzyme expressed in all mammals except humans. In mice, this sugar is synthesized in several somatic tissues. Expression of NeuGc-GM3 ganglioside was reached by in vitro incubation with NeuGc-rich bovine submaxillary mucin and evaluated by slot-blot and immunohistochemistry assays using the 14F7 antibody.
Tumor cells treated with mucin or purified NeuGc were injected s. In vitro incubation of these NeuGc-negative cells with NeuGc-rich mucin increased the presence of NeuGc in cell membranes for at least h, as a component of the GM3 ganglioside. Preincubation with NeuGc-rich mucin reduced tumor latency and increased the metastatic potential of tumor cells in syngeneic animals. Similar results were obtained when cells were incubated with purified NeuGc alone. Conclusion Our results indicate that B16 and F3II mouse tumor cell lines do not express NeuGc in cell membranes but they are able to incorporate NeuGc from an exogenous source, contributing to the malignant phenotype of melanoma and mammary carcinoma cells.
As an alternative source of organs for transplantation into humans, attention has been directed to pigs due to their similarities in biological features and organ size. However, severe immune rejection has prevented successful xenotransplantation using pig organs and tissues. To overcome immune rejection, recently developed genetic engineering systems such as TALEN coupled with somatic cell nuclear transfer SCNT to make embryos could be used to produce pigs compatible with xenotransplantation.
Cloned embryos using 3 different genetically modified cell clones were respectively transferred into 3 recipients, with Successful genetic disruption of the CMAH gene was confirmed by sequencing, showing lack of expression of CMAH in tail-derived fibroblasts of the cloned piglets.
Besides decreased expression of Neu 5 Gc in piglets produced by SCNT, antibody-mediated complement-dependent cytotoxicity assays and natural antibody binding for examining immuno-reactivity of the quadruple gene modified pigs derived from endothelial cells and fibroblasts were reduced significantly compared to those of wild type animals. We conclude that by combining the TALEN system and transgenic cells, targeting of multiple genes could be useful for generating organs for xenotransplantation.

Aspartic acid helps every cell in the body work. Although acid rain is fading as a political issue in the United States and funds for research in this area have largely disappeared, the acidity of rain in the Eastern United States has not changed significantly over the last decade, and it continues to be a serious environmental problem. Acid deposition commonly called acid rain is a term applied to all forms of atmospheric deposition of acidic substances - rain, snow, fog, acidic dry particulates, aerosols, and acid -forming gases.
Water in the atmosphere reacts with certain atmospheric gases to become acidic.
n-glycolylneuraminic acid neu5gc: Topics by theranchhands.com
For example, water reacts with carbon dioxide in the atmosphere to produce a solution with a pH of about 5. These acid -producing gases are released to the atmosphere through natural processes, such as volcanic emissions, lightning, forest fires, and decay of organic matter. Accordingly, precipitation is slightly acidic , with a pH of 5. In industrialized areas, most of the acid -producing gases are released to the atmosphere from burning fossil fuels. Major emitters of acid -producing gases include power plants, industrial operations, and motor vehicles.
Acid -producing gases can be transported through the atmosphere for hundreds of miles before being converted to acids and deposited as acid rain. Because acids tend to build up in the atmosphere between storms, the most acidic rain falls at the beginning of a storm, and as the rain continues, the acids "wash out" of the atmosphere. Such a precipitation is prevented by dissolving in the acid solution an amount of 5-sulfosalicylic acid which is at least sufficient to sequester significant proportions of ferric ions when the pH of the acid is from 0. The amount of the 5-sulfosalicylic acid may be from 0. Ibotenic acid and thioibotenic acid. In this study, we have determined and compared the pharmacological profiles of ibotenic acid and its isothiazole analogue thioibotenic acid at native rat ionotropic glutamate iGlu receptors and at recombinant rat metabotropic glutamate mGlu receptors expressed in mammalian cell lines Thioibotenic acid has a distinct pharmacological profile at group III mGlu receptors compared with the closely structurally related ibotenic acid ; the former is a potent low microm agonist, whereas the latter is inactive.
By comparing the conformational energy profiles of ibotenic and thioibotenic acid The production of formic acid by the oxidation of methane with a metallic oxide or by the interaction of carbon monoxide and water vapor in the presence of a catalyst, preferably a metallic oxide, is described along with the destructive distillation of carbonaceous material in the preesnce of formic acid vapor for the purpose of increasing the yield of condensible hydrocarbons.

Acid deposition, commonly known as acid rain, occurs when emissions from the combustion of fossil fuels and other industrial processes undergo complex chemical reactions in the atmosphere and fall to the earth as wet deposition rain, snow, cloud, fog or dry deposition dry particles, gas. Rain and snow are already naturally acidic , but are only considered problematic when less than a ph of 5. When these two compounds react with water, oxygen, and sunlight in the atmosphere, the result is sulfuric H 2 SO 4 and nitric acids HNO 3 , the primary agents of acid deposition which mainly produced from the combustion of fossil fuel and from petroleum refinery.
Airborne chemicals can travel long distances from their sources and can therefore affect ecosystems over broad regional scales and in locations far from the sources of emissions. When scientists learned that acid rain could harm fish, fear of damage to our natural environment from acid rain concerned the American public. Research by USGS scientists and other groups began to show that the processes resulting in acid rain are very complex.
Scientists were puzzled by the fact that in some cases it was difficult to demonstrate that the pollution from automobiles and factories was causing streams or lakes to become more acidic. Further experiments showed how the natural ability of many soils to neutralize acids would reduce the effects of acid rain in some locations--at least as long as the neutralizing ability lasted Young, The USGS has played a key role in establishing and maintaining the only nationwide network of acid rain monitoring stations.
The USGS supports about 72 of these sites. The reported increasing acidity of rainfall raises many interesting ecological and chemical questions.
In spite of extensive studies in Europe and North America there are, for example, great uncertainties in the relative contributions of strong and weak acids to the acid -base properties of rainwater. Unravelling this and similar problems may require even more rigorous sample collection and analytical procedures than previously employed. Careful analysis of titration curves permits inferences to be made regarding chemical composition, the possible response of rainwater to further inputs of acidic components to the atmosphere, and the behavior to be expected when rainwater interacts with the buffers present in biological materials and natural waters.
